Marc PRUDHOMME : "Acoustofluidic sensor with functionalized bubbles for the detection of bioanalytes"

Abstract :  The development of microfluidic in the early XXth century has lead to the growth of compact, integrated and user-friendly biosensors. These devices aim at detecting and often quantifying a biological element called an analyte (virus, bacteria, protein, cell...) evolving in biological fluids such as the blood, the saliva or the urin. However, most of these biosensors share a common inconvenient: they are mostly disposable. Indeed, the regenration of the capture surface, flat and fixed within the device, is complicated or even impossible to proceed,
due to the strong chemical bonds between the surface and the receptors. This thesis consists in transferring the capture surface to a mobile and easily regeneration surface within the device. The microbubbles are widely studied in digital microfluidic, due to their singular acoustic behavior. They are behind major breakthroughs in the biomedical domains, especially in imaging and therapy. The goal of this thesis is to use the bubbles surface generated in a microfluidic chip as capture interface. The microfluidic sensor will integrate the following steps: generation and functionalisation of the microbubbles surface, static organisation of the bubbles inside a chamber, injection of the analytes solution and capture at surface. The couple biotin/streptavidin is used in order to demonstrate the efficiency of the sensor. Besides, the capture is assesed by acoustic probing, in order to have a sensitive, label free and integrated detection method.[...]
